Does IFS Cloud 22R2 support ‘V4 Sign Authentication method’?

NOTE:
This is also known as ‘AWS Signature V4’ and the REST call is from IFS to a third party.

No, IFS Cloud does not support it, but fortunately, for Amazon SP-API, AWS Signature is not needed anymore, it works with Oauth2
