Is it possible to uncheck “use hardware accelerated graphics” for a base profile?

No. This is a device specific setting. Means that you can have two unique setup in two devices - (ie. your laptop and desktop).

The setting is saved in IsolatedStorage - Its in windows drive (C:) - C:\Users\rudhlk\AppData\Local\IsolatedStorage

It would be a binary file having .txt extension (developer_settings.txt).
